Product Details

The Investigator Argus X-12 QS Kit is a multiplex solution for human identification. The 12 X-chromosomal markers, D21S11 and Amelogenin are co-amplified. The autosomal marker, D21S11, enables alignment of the Investigator Argus X-12 QS Kit profile with a profile from any other autosomal STR kit from the CODIS or ESS market, to minimize the risk of sample mix-ups. The kit utilizes QIAGEN's fast-cycling technology, enabling amplification in approximately 80 minutes. Highly sensitive, with rapid and reliable results from trace or reference DNA, the Investigator Argus X-12 QS Kit is suitable for paternity or kinship testing and for gonosomal STR analysis of forensic stains.

The Investigator Argus Y-12 QS Kit is a versatile tool for analyzing Y-chromosomal markers. It is intended for the reliable generation of male DNA profiles from mixtures of male and female DNA without the necessity of separating sperm and female cells. Furthermore, it excellently complements the Investigator Argus X-12 QS Kit for paternity testing.

Both kits includes an internal control (Quality Sensor) that provides helpful information on PCR efficiency and the presence of PCR inhibitors in the tested samples. The genetic analysis is calibrated with BT5 Matrix.

Investigator Argus QS Kits meet ISO 18385 requirements.

Performance

The Investigator Argus X-12 QS Kit utilizes fast-cycling technology to provide reliable results in approximately 80 minutes.

The Fast Reaction Mix 2.0 is based on proven QIAGEN Multiplex PCR technology that has been further optimized for direct amplification. Assay performance tests in the presence of increasing concentrations of inhibitors showed superior inhibitor resistance. This means reliable, high-quality results from casework samples, but also from direct amplification of typical reference samples such as blood or epithelial cells on FTA paper.

The Investigator Argus X-12 QS Kit was validated according to ENFSI and SWGDAM guidelines.

The Investigator Argus Y-12 QS Kit allows the generation of male DNA profiles from mixtures of male and female DNA up to a ratio of 1:4000 so that separation of sperm from female cells or differential lysis is not required.

Generation of DNA profiles using the Investigator Argus Y-12 QS Kit conforms to the guidelines of the International Society for Forensic Genetics. The kit was validated and evaluated using the GeneAmp 9700 thermal cycler and the ABI PRISM 310, ABI PRISM 3100 and Applied Biosystems 3130 Genetic Analyzers.

For both kits, the optimal amount of DNA under standard conditions is 0.2–0.5 ng. Internal validations demonstrated reliable results with <0.1 ng DNA.

Principle

The Investigator Argus X-12 QS Kit uses QIAGEN's fast-cycling PCR technology for the simultaneous amplification of 12 X-chromosomal markers, D21S11 and Amelogenin. Amplification of the 12 X-chromosomal markers (see table “Investigator Argus X-12 QS Kit dyes and corresponding markers”), with heightened discrimination power, meets the needs for complex kinship and paternity testing and helps fulfill the demands of complicated deficiency cases involving at least one female. The 12 X-chromosomal markers are clustered into four linkage groups (three markers per group; see table “Linkage groups”), and thus each set of three markers is handled as a haplotype for genotyping.

The additional autosomal marker D12S11 acts as alignment marker. Now, you have the possibility to align your X-chromosomal profile from the Investigator Argus X-12 QS Kit with a profile of any other autosomal STR kit from the expanded CODIS or ESS market, to minimize the risk of sample mix-ups.

The Investigator Argus Y-12 QS Kit is a multiplex application that amplifies 12 Y-chromosomal STR loci, specifically of the so-called extended minimal haplotype (see tables “Investigator Argus Y-12 QS Kit dyes and corresponding markers” and "Chromosomal mapping of the Investigator Argus Y-12 QS Kit"), as recommended by the Scientific Working Group on DNA Analysis Methods (SWGDAM). Developed for use with mixed DNA samples, the Investigator Argus Y-12 QS Kit generates reliable male DNA profiles from DNA mixtures with male to female ratios of up to 1:4000. This makes it unnecessary to separate sperm from female cells or run differential lysis.

The innovative Quality Sensors included in Investigator Argus QS Kits provide additional information on the sample quality and PCR success beyond the profile of your sample. With the Quality Sensor (QS) included as an amplification control, it is possible to determine:

  • A successful PCR run
  • A failed PCR run
  • Lack of DNA in your sample

This additional information will help you analyze your data, and guides you with respect to further sample processing. It will also help you reduce unnecessary PCR re-runs, saving you time and money.

Procedure

The 12 X-chromosomal markers, D21S11 and the gender-specific Amelogenin, are amplified simultaneously in a single PCR run and labeled with one of the dyes listed in table "Investigator Argus X-12 QS Kit dyes and corresponding markers". The 12 polymorphic Y-chromosomal loci are simultaneously amplified in a single PCR and labeled with the dyes listed in the table "Investigator Argus Y-12 QS Kit dyes and corresponding markers".

The amplified loci undergo capillary electrophoresis. The results can be analyzed using dedicated software, such Applied Biosystems GeneMapper ID-X Software. See the table "Technical specifications" for more information.

Applications

The Investigator Argus X-12 QS Kit enables simultaneous amplification of 12 X-chromosomal markers, and is highly suitable for kinship and paternity testing, especially deficiency cases, population genetics and anthropological studies and analysis of stains with female traces on a male background.

The most useful applications for the Investigator Argus X–12 QS Kit include:

  • Paternity with related putative fathers (superiority of ChrX-markers if father/son)
  • Paternity testing for daughters (only ChrX of males is transmitted to all females)
  • Motherhood (verification of mother/son)
  • Sisterhood (identical paternal ChrX-haplotype)
  • Deficiency cases with missing putative father (reconstruction of family trees)

The Investigator Argus Y-12 QS Kit combines the inherent male specificity of Y-STRs with robust performance. It enables simultaneous amplification of 12 Y-chromosomal STR loci, and is highly suitable for kinship and paternity testing, analysis of sexual assault samples, analysis of forensic stains and samples and generating male DNA profiles from mixed samples.

The most useful applications for the Investigator Argus Y–12 QS Kit include:

  • Sexual assault sample analysis
  • Analysis of trace quantities of male DNA present in a large background of female DNA
