Multi-Analyte ELISArray Kits are the simplest simultaneous multi-analyte ELISA available. Highly specific capture antibodies for key cytokines and chemokines are arrayed in a 96-well plate. One easy ELISA protocol allows the detection of 12 cytokines or chemokines simultaneously in multiple samples.

ELISArray protocol.

Multi-Analyte ELISArray Kits follow a traditional ELISA protocol that can be completed in any laboratory with a microplate reader.

Multi-Analyte ELISArray plate layout.

Each Multi-Analyte ELISArray contains twelve 8-well strips (represented by colored columns) coated with a specific capture antibody for a cytokine or chemokine. Six samples can be analyzed at one time, in addition to controls.

Patterns of Th1/Th2 cytokine induction.

Th1/Th2 cytokines were induced by human peripheral blood mononuclear cells in response to PMA (50 µg/ml) and ionomycin (1 µg/ml). Time-dependent patterns of induced cytokines and relative cytokine amounts were profiled and plotted.

Optimized ELISA conditions enable the same development or incubation time.

Similar standard curves are observed for all optimized cytokine and chemokine assays in the Multi-Analyte ELISArray Kits. All standard curves are virtually superimposable indicating that all 12 assays provide similar linearity and sensitivity under the same standardized conditions and development or incubation times.

Performance

One example of a Multi-Analyte ELISArray Kit application is as follows. A Multi-Analyte ELISArray Kit was used to profile multiple cytokines secreted by stimulated human peripheral blood mononuclear cells (PBMCs). The time-dependent (0, 6, 18, 24, and 48 hours) patterns of 8 Th1/Th2 cytokine inductions by human PBMCs were montored in response to PMA (50 µg/ml) and ionomycin (1 µg/ml) (see figure "Patterns of Th1/Th2 cytokine induction". The relative amount of each of the 8 cytokines was also profiled using the Multi-Analyte ELISArray Kit.

Large amounts of IL-2 and IFNγ, and to a lesser extent TNFα, are induced throughout, and apparently beyond, the time course tested. Only more moderate amounts of IL4 and IL5 are induced, reaching a maximum at approximately 24 hours of stimulation. The even lower levels of IL10 and IL13 that are secreted also reach a maximum at 24 hours and appear to decrease again at later times. The amount of IL12 is not detectable after 48 hours of constant stimulation.

Principle

Multi-Analyte ELISArray Kits profile 12 cytokines in 6 cell supernatant (media), serum, or plasma samples using one simple protocol and one standardized development time (see figure "Multi-Analyte ELISArray plate layout"). Multi-Analyte ELISArray Kits provide researchers with a rapid assessment of the level of multiple proteins (cytokines and chemokines) in multiple samples.

Commercially available capture and detection antibodies are screened to achieve the highest possible sensitivity and linearity for each cytokine or chemokine assay. All assays are optimized under uniform conditions to allow simultaneous detection without compromising performance (see figure "Optimized ELISA conditions enable the same development or incubation time").

Procedure

Multi-Analyte ELISArray Kits use a standard sandwich ELISA protocol (see flowchart "ELISArray protocol"). There is no requirement for special equipment; kits are compatible with a standard ELISA plate reader.

Applications

Multi-Analyte ELISArray Kits are highly suited for research into autoimmune and immune disorders, cancer, immunology, infectious diseases, and many other areas of biological and medical research. They can be used in any application typically analyzed using individual ELISA kits.

Inflammatory Cytokines Multi-Analyte ELISArray Kits
Introduction
The Mouse Inflammatory Cytokines & Chemokines Multi-Analyte ELISArray Kit analyzes a panel of 12 pro-inflammatory cytokines using a conventional ELISA protocol all at once under uniform conditions. The cytokines & chemokines represented by this array are IL1A, IL1B, IL2, IL4, IL6, IL10, IL12, IL17A, IFN, TNF, G-CSF, and GM-CSF. The pro-inflammatory cytokines IL1, IL6, and TNF are also endogenous pyrogens. IL4 and IL10 can also act as anti-inflammatory cytokines depending on their concentrations. The complete kit features the best possible combination of capture and detection antibodies for each protein in the panel and all of the needed colorimetric detection reagents. Using the same ELISA protocol and development or incubation time, you can easily profile the levels of a focused panel of 12 proteins related to inflammation with this array.
