This novel kit extracts extracellular and intracellular microbial protein without co-extracting humic substances or other interfering compounds. The bead beating protocol uses a mixture of glass and ceramic beads along with two buffers to efficiently break down intracellular and extracellular protein. All materials are certified protein- and protease-free.

The resulting protein pellet has fewer impurities compared to other extraction methods and can be easily resuspended for further analysis or storage. Proteins are ready for 1D and 2D polyacrylamide gel electrophoresis and mass spectrometry (2D LC-MS/MS).

The NoviPure Soil Protein Kit was previously sold by MO BIO under the same name.

Challenging soil types still yield high amounts of protein using the NoviPure Soil Protein Kit. The kit was used to extract total protein from 5 g samples of sterile soil and sediments spiked with E. coli. To visualize protein results, 20 µl of each sample was processed using a stain-free 1D SDS-PAGE gel. In all cases, protein bands were clear and matched the E. coli culture control.

Protein was extracted using the NoviPure Soil Protein Kit and analyzed by 2D LC-MS/MS, where total ion chromatogram data reveals base peaks of protein. Visualized here is fraction 4 of the SCX cation exchange. The NoviPure Soil Protein Kit extracted protein from 5 g of sterile compost samples spiked with E. coli. Digestion and peptide enrichment followed using reverse phase ion exchange. Comparing raw spectra results with the SwissProt protein database (March 2013 release), 1076 unique proteins were identified.

C. albicans was added to 5 g of sterile compost samples. Protein was then extracted from those samples. Samples processed using the NoviPure Soil Protein Kit show greater protein extraction compared to the sucrose and phenol methods.
