Product Details

Investigator IDplex Kits are multiplex STR assays for human identification covering the 13 CODIS (Combined DNA Index System) core loci, D2S1338, D19S433 and Amelogenin.

The kits utilize our fast-cycling technology, enabling amplification in just 45–90 minutes, depending on the kit used. Highly sensitive with rapid and reliable results from trace DNA, the Investigator IDplex Plus Kit is suitable for all forensic applications and paternity testing. With rapid and reliable results from reference samples such as buccal swabs, as well as buccal cells or blood on papers, the Investigator IDplex GO! Kit is suitable for reference testing in forensic applications and paternity testing. The genetic analyzer is calibrated with BT5 Matrix.

Investigator IDplex Kits meet ISO 18385 requirements.

Performance

Investigator IDplex Kits utilize fast-cycling technology to provide reliable results in STR assays in just 45 minutes for reference samples with direct amplification or 90 minutes for any other sample.

The Fast Reaction Mix is based on proven QIAGEN multiplex PCR technology that has been optimized to increase inhibitor tolerance. Assay performance tested in the presence of increasing concentrations of 6 types of inhibitor (humic acid, hematin, tannic acid, indigo carmine, calcium and collagen) shows high sensitivity and superior inhibitor resistance. This enables reliable, high-quality results from casework samples, but also directly from reference samples such as blood or epithelial cells on FTA paper.

Investigator IDplex Kits were validated and evaluated using the GeneAmp 9700 thermal cycler and the Applied Biosystems 3500 Genetic Analyzer.

The recommended amount of DNA under standard conditions for use with the Investigator IDplex Plus Kit is 0.5 ng. Internal validations demonstrated robust and balanced results with 0.2–2 ng DNA, and reliable results with <0.1 ng DNA.

Principle

Investigator IDplex Kits use QIAGEN's fast-cycling PCR technology for the rapid, simultaneous amplification of the 13 CODIS (Combined DNA Index System) core loci, D2S1338, D19S433 and Amelogenin. Carefully designed primers reduce overlap of loci alleles, thereby minimizing the likelihood of misinterpretation.

Procedure

The 15 polymorphic autosomal STR markers (D2S1338, D3S1358, D5S818, D7S820, D8S1179, D13S317, D16S539, D18S51, D19S433, D21S11, CSF1PO, FGA, TH01, TPOX and vWA), and the gender-specific Amelogenin are amplified simultaneously in a single PCR and labeled with one of the dyes listed in the table "Investigator IDplex Kit dyes and corresponding markers".

Investigator IDplex Kit dyes and corresponding markers

Dye Marker
6-FAM Amelogenin TH01 D3S1358 vWA D21S11
BTG TPOX D7S820 D19S433 D5S818 D2S1338
BTY D16S539 CSF1PO D13S317 FGA
BTR D18S51 D8S1179

The amplified loci then undergo capillary electrophoresis. The results can be analyzed using dedicated software, such as GeneMapper ID-X. See the table "Technical specifications" for more information.

Applications

Investigator IDplex Kits are for rapid and reliable generation of DNA profiles from blood, buccal swabs and forensic stains or reference samples and are highly suited for:

  • Criminal database, forensic reference or casework samples
  • Paternity testing
  • Population genetics studies
  • Other human identity testing applications
